Development and characterization of new dairy products with increased fiber and protein content

The bachelor thesis was focused on the production and characterization of new dairy products with increased protein and fiber content. Ground wheat bran was used as the fiber source and wheat bran protein extract was used as the protein source. From an ecological point of view, it would be ideal if 100% of the raw material was used and nothing was left as waste. We tried to approach this by using the waste (by-product) that arises during the production of flour as an additive to dairy products. The theoretical part focused on the characterization of fermented and unfermented dairy products and their production, the nutritional value of proteins and fiber and the composition and benefits for human health of wheat bran, protein isolation from wheat bran and common analytical methods to determine rheological properties, viable cell numbers. water binding and sensory acceptability. Two basic dairy products were created, one fermented and one unfermented, and subsequently fortified with a source of fiber (wheat bran) and a source of protein (protein concentrate from wheat bran). All products were subsequently characterized for characterization. Water binding, rheological and sensory properties were determined. The fermented dairy product was also subjected to cytometric analysis to determine the number of viable cells. The experimental part describes the methods we use to prepare and characterize new products. After evaluating the measured data, options for optimizing the acceptability of products by consumers were proposed. The results showed that the organoleptic properties were negatively affected by the addition of wheat bran and protein concentrate.
The characterisation of selected type of „bio“ dairy product

This bachelor thesis deals with the sensory evaluation of kefir. Specifically the differences in the sensory properties of kefir produced using the same method from various types of milk were observed. The theoretical part was focused on kefir, its characteristics, composition and production using kefir grains. The experimental part deals with the comparison of four kefir samples based on sensory evaluation. The evaluation was made by the students from Faculty of Chemistry BUT, the untrained assessors. Therefore the evaluation results may be closely reflecting the opinions of common consumers. The samples of kefir were subjected to sensory evaluation by the ranking test and the pleasantness of chosen properties (appearance, colour, texture, odour and taste) was evaluated using a seven-point scale, as well as the overal acceptability of samples. The results were statistically processed and graphically evaluated.
Detection of probiotic bacteria in milk diary food products using PCR technique

Probiotic bacteria play an important role in a healthy lifestyle. They help the consumer maintain the balance of intestinal microflora and prevent the overgrowth of harmful bacteria. Accurate identification and characterization of these probiotic strains is essential for research and the food industry. For exact identification, the use of molecular biological methods is necessary, thanks to which it is possible to validate probiotic products for commercial use. In this bachelor thesis, the DNA of probiotic bacteria was isolated from available dairy products. Two methods of isolation were used to isolate bacterial DNA, both of them provided sufficiently concentrated and high-quality DNA for further analysis by polymerase chain reaction (PCR). The presence of the Bacteria domain, genera Lactobacillus and Bifidobacterium were proved. Finally, the presence of Lactobacillus acidophilus species was also detected in the products.
Microbiological analytical methods suitable for dairy industry

The theoretical part of the thesis is focused on probiotics in dairy products. Thesis deals with molecular genetic methods, which are used to analyze DNA. Especially are discussed methods used for isolation bacterial cells belonging to genus Lactobacillus. The polymerase chain reaction (PCR) is the basic technique at present time. Short chain of oligonucleotide primers are used to amplification specific parts of DNA molecule chain. The practical part of the thesis is focused on the DNA from pure bacterial culture and probiotic dairy product. DNA was isolated using methods of phenol-chloroform extraction and magnetic separation. For magnetic separation was used magnetics particles covered with carbonyl functional groups. Quality of the DNA was confirmed by PCR amplification. Primers specific for domain Bacteria and genus Lactobacillus and Bifidobacterium was used.
Probiotic bacteria and authenticity of milk products

Probiotics are living microorganisms that have positive effects on human health after consumption. The theoretical part of the bachelor thesis describes properties and health effects of probiotics and their use in food industry. The experimental part was focused on the isolation of PCR-ready DNA from two dairy probiotic products. The presence of target bacterial DNA was confirmed using PCR methods.
Isolation of PCR-ready DNA from dairy products for baby nutrition

The work was focused on isolation of PCR-ready DNA and the identification of probiotic lactic acid bacteria that were isolated from five milk product for infant nutrition. DNA was isolated from crude cell-lysates of the products by magnetic P(HEMA-co-GMA) microspheres. DNAs isolated from crude cell lysates of control strains using phenol extraction method were used as positive controls. Using PCRs DNA of genera Bifidobacterium and species B. animalis, B. bifidum, B. breve, B. infantis, B. longum and Streptococcus thermophilus species were identified in products. The results obtained are consistent with the data declared by the manufacturers.
Identification of selected species of lactic acid bacteria in dairy products

Lactic acid bacteria are natural part of the human gastrointestinal tract. They are often used in food supplements and for the production of fermented dairy products. This thesis focuses on the identification of selected species of lactic acid bacteria and bifidobacteria in cheese and dairy products. Bacterial DNA was isolated by magnetic particles P(HEMA-co-GMA) from crude cell lysates from 9 products. Isolated DNA was amplified in genus-specific and species-specific polymerase chain reactions (PCR). The obtained amplicons were detected by agarose gel electrophoresis. The results of PCR were compared with those provided by the manufacturers and there has been declared a match.

The theoretical part of diploma thesis is focused on summarizing current knowledge about S. aureus, its pathogenesis, occurrence, and possibilities of its detection in milk and dairy products. The experimental part of diploma thesis is aimed at determining S. aureus using various cultivation methods. S. aureus was determined using B-P agar with the addition of RPF, ClearMilku test, MicroFast®, and PetrifilmTM. The determination was evaluated as follows: B-P agar with the addition of RPF > PetrifilmTM > MicroFast® > ClearMilk test. Another part of the experimental diploma thesis deals with the possibility of detecting S. aureus using a newly developed biosensor detection system.

This bachelor thesis was elaborated in 2019 and 2020 at the Department of Post-Harvest Technology of Horticultural Products of the Faculty of Horticulture, Mendel University in Brno. Bachelor thesis deals with the division of microorganisms, description of their nutrition and reproduction, important genera in terms of both positive and negative. The influence of microorganisms for food industry and important fermentation processes for beverage industry is described. All important microorganisms for the production of wine, beer, spirits and milk fermented beverages are included. The work also follows the microorganisms, which cause various defects in the bev-erage.
Microbiology of raw milk and selected dairy products

Milk is an important part of the human diet due to its composition. The aim of the bachelor thesis was to create a review focused on the microbiota of raw cow's milk and dairy products. The thesis summarizes the ways of primary and secondary microbial contamination of milk and describes the intrinsic and extrinsic factors influencing the growth of microorganisms. A particular attention is focused on mastitis, which are the main source of primary contamination of milk. The great emphasis is attended to the necessity of strict following of hygiene regulations during manipulation with this raw material. In the last part, potential health risks associated with consumption of milk and dairy products is mentioned. .
